Mathews was born in Melbourne, but has spent most of her life in Sydney. She was offered a scholarship at The University of the Arts in Philadelphia, Pennsylvania, U.S., which she completed. She then enrolled in and completed a two year acting course in Surry Hills, Sydney, where she met fellow Home and Away actor Jon Sivewright, who plays Tony Holden, her husband on the show.

Mathews played guest roles in Seven Network series, such as Always Greener, All Saints and Blue Heelers.

In 2006, Mathews joined the cast of the long-running Australian soap opera Home and Away, as Rachel Armstrong, a psychiatrist (later the main doctor in A&E). Mathews received a Logie Award for Most Popular New Female Talent in 2007.

Mathews played a nurse in Gabriel (2007).
